The influence of the dermis on the epidermis during aging

Cellular communication participates in maintaining the skin's biological functions and contributes to its beauty. SILAB got particularly interested in transcompartmental communication between the dermis and the epidermis during aging, an essential component to ensure skin longevity.

To this end, a novel in vitro modeling study was implemented. First, a proteomic study on young fibroblasts revealed that 38% of proteins secreted are involved in an intercellular communication function. SILAB highlighted that, in the course of aging, the synthesis of more than half of proteins involved in cell communication from the dermis to the epidermis is altered. Moreover, miRNA expression (epigenetic markers) transported by extracellular vesicles (including exosomes) is also decreased. Finally, it was demonstrated that this communication shortfall has a significant impact on the epidermal biological functions, in particular a reduction of keratinocyte proliferation and differentiation, of hydration and cell cohesion, as well as a weakened anchoring of the dermal-epidermal junction.

A global anti-aging action

Given this observation, SILAB has developed COMMUSYS® , an active ingredient capable of regulating the molecular players involved in communication from the dermis to the epidermis, thereby reactivating epidermal functions. Indeed, the secretome of aged fibroblasts treated with 0.4% COMMUSYS® increases keratinocyte processes such as cell proliferation and cohesion, keratinocyte differentiation, hydration and anchoring of keratinocytes at the dermal-epidermal junction.

These results translate on a mature Caucasian panel into positive effects on epidermal renewal and thickness, assessed using optical biopsies. As of 14 days, COMMUSYS® at 1.7% significantly increases the water content of the epidermis (+7.2%), smooths the microrelief (density of surface irregularities: -14.2%) and improves complexion radiance (reflection: +12.6%). These effects continue and intensify after 28 days. An improvement of the hydration level and complexion radiance was also observed in Asian subjects after 28 and 56 days of use.
